RNA Sequencing, Elevate (Cloudbreak Freestyle™)

Cloudbreak Freestyle chemistry was used to perform Elevate RNA sequencing (RNA-Seq) on human reference RNA samples.

Methods

Libraries were prepared using the RNA Library Prep Kit (#830-00019) and 100 ng of Universal Human Reference RNA (Thermo Fisher). Enrichment for mRNA was accomplished via the Elevate mRNA Capture Kit for RNA Library Prep (#830-00021) followed by enzymatic shearing, ligation of adapters, size selection, and addition of indexing primers using the Elevate RNA Index and Adapter Kit (#830-00020). Libraries were sequenced via Cloudbreak Freestyle on the AVITI™ System with a 2 x 75 read length. Bases2Fastq was used to generate FASTQ files.
